DURHAM, N.C. A three-dimensional image of one of the proteins that serves as an on-off switch as it binds to receptors on the surface of a cell suggests there may be a sort of main power switch that could be tripped. These surface receptors are responsible for helping cells discern light, set the heart racing, or detect pain.

The finding, published online April 21, 2013, in the journal Nature by a research collaboration involving this year's Nobel laureates in chemistry, could help in the development of more effective drugs to switch on or off the cell receptors that regulate nearly every bodily function. Already, up to half of all drugs engage these receptors, including antihistamines and beta blockers, but many of the intricacies of how these important proteins work remain unknown.

"It's important to understand how this extraordinary family of receptors work," said co-author Robert J. Lefkowitz, M.D., James B. Duke Professor of Medicine and Howard Hughes Medical Institute Investigator. "This is the kind of finding that answers a basic curiosity, but can also be of benefit if we can develop new drugs or improve the ones we have."

The research marks a collaborative reunion between Lefkowitz and Brian K. Kobilka, M.D., chair of molecular and cellular physiology at Stanford University School of Medicine. The two researchers friends who first collaborated when Kobilka was a trainee in Lefkowitz's laboratory at Duke - shared the 2012 Nobel Prize in Chemistry for their discoveries involving the G-protein coupled receptors (GPCRs), which are activated by signaling proteins to detect hormones, neurotransmitters, pain, light.

In the current work, the researchers used X-ray crystallography to develop an atom-scale image of one of the principal signaling molecules that regulate GPCRs. This protein is called beta-arrestin1, which, among other things, works to dim a cell's response to hormones such as adrenalin.

The researchers were able to isolate and capture the beta-arrestin1 protein in an active state as it binds to a segment of the GPCR a first. That snapshot, in high resolution, revealed that the structural conformation or shape of the protein in its active state is strikingly different than when it is inactive.

Such changes suggest there may be a general molecular mechanism that activates the beta-arrestin1 a sort of main switch that controls the multi-functional signaling proteins.

"It's like there are brakes on in beta-arrestin1, and then when the beta-arrestin1 binds to a GPCR, the brakes are released, thereby activating beta-arrestin1," said Arun K. Shukla, PhD, assistant professor of medicine at Duke and co-lead author of the study.

The researchers are now pursuing additional structural imaging of the signaling complex consisting of beta-arrestin1 and the entire receptor protein.

No need for new sanctions measures on Iran: UK's Hague

LUXEMBOURG (Reuters) - The European Union should focus on implementing its current sanctions against Iran over Tehran's nuclear program rather than imposing new measures for the moment, Britain's Foreign Secretary William Hague said on Monday.

Hague was speaking days after U.S. Secretary of State John Kerry asked for patience from U.S. senators pressing for tougher sanctions over Tehran's nuclear ambitions, saying there is uncertainty in Iran two months before its June 14 election.

Hague said economic pressure from Europe was increasing on the Islamic Republic, thanks to wide sanctions against banking and industry imposed on Iran last year, and there was time to wait with more.

"We must maintain (current) sanctions pressure, not proposing new ones," he told reporters ahead of a meeting of EU foreign ministers in Luxembourg.

Western governments hope economic pressure will persuade Tehran to scale back its most sensitive nuclear work, which they fear has a covert military goal.

Antares Rocket Launch Is A Success, In Test Of Orbital Supply Vehicle

The Antares rocket launch is back on for 5 p.m. ET Sunday afternoon, as engineers and spectators look for the rocket to lift off from a launch pad at NASA's Wallops Flight Facility. A check of all systems at 10 minutes before its launch was positive.

Update at 5:10 p.m. ET. Orbit Achieved, Payload Separated:

Antares moved smoothly through its stages and achieved orbit, drawing applause from engineers watching in the control room. Soon after, it delivered its practice payload into orbit around the Earth. The test mission has about eight more minutes remaining.

Update at 5:02 p.m. ET. Liftoff Is A Success:

The Antares rocket soared above Wallops Island Sunday afternoon. Just before the engines were throttled down in Stage 1, the vehicle was traveling at more than 7,000 miles per hour. Our original post continues:

The mission had been postponed twice this week, after a connecting cable came loose on Wednesday and unfavorable winds forced a one-day delay on Saturday. Officials believe the launch may be visible from sites all along the eastern U.S. coast.

Around 3:30 p.m. ET Sunday, the rocket was approved for launch and fueling of the vehicle began, NASA reported.

The NASA facility is located just behind the beach on Wallops Island on Virginia's Eastern Shore. Antares is a product of a joint development project between NASA and Orbital Sciences Corp., based in Virginia.

As NPR's Geoff Brumfiel reported Wednesday, a successful test would eventually make Orbital "the second private company after California's SpaceX to dock with the space station."

PFT: Colts raise over $650K for cancer research

With the NFL Draft approaching, we?re taking a team-by-team look at the needs of each club. Up next is the team with the No. 30 overall selection, the Atlanta Falcons. They have a total of 11 picks, and aren?t afraid to make bold moves, so their picking somewhere north of 30 is a real possibility.

Defensive end: There?s already noise about the Falcons wanting to move up in the order, and it makes sense that their target would be a pass-rusher.

John Abraham was still producing right up until the time they released him, and they have to find someone to replace that production other than Osi Umenyiora, who has not produced at the same consistent level. If they can get into the top half of the first round, they can find someone in the Abraham mold, before the run starts.

Cornerback: This would be the other drastic need that could be the target for a move-up. It seems like forever ago they were three-deep with excellent players. But with Brent Grimes gone to Miami and and Dunta Robinson released and resurfaced in KC, they?re down to Asante Samuel and a bunch of guys.

Linebacker: You could probably run down the list of defensive positions, and the Falcons could stand to upgrade. Sean Weatherspoon is quite good, but the rest of their linebacking corps is fairly ordinary.

Tackle: The thinking is the release of Tyson Clabo opened the door for Lamar Holmes to start at right tackle, and that could work. But they still need cover here, particularly if newly rich left tackle Sam Baker goes back to the 2011 version instead of the 2012 salary-push version.

Guard: The retirement of Todd McClure leaves a big hole, but they could slide 2012 second-rounder Peter Konz over from right guard. But that leaves another vacancy. They have some in-house candidates, but need depth here.

As good as they are, they?re not as well-covered as you?d think. They have nothing to speak of in terms of depth on defense. Heck, they don?t even have a full complement of starting-caliber players on that side of the ball.

But their offensive skill-position talent is so good, it might not matter. As long as Matt Ryan, Steven Jackson, Julio Jones, Roddy White and Tony Gonzalez are together, the Falcons are going to be among the best in the NFC.

But until they get at least a little bit better on defense, it?s going to be hard for them to make the next step.

Their offseason efforts have focused on retaining their own, and making a few surgical signings of veterans who came looking for rings. Their team is good enough that 11 draft picks aren?t making their 53-man roster, so they might as well make some moves, and see if they can plug some starters in on defense.
